- St. James City is the largest area, with over half of the island's population. Most of the homes are located along direct access canals that lead to the Gulf of Mexico. Colorful waterside restaurants with boat access and watering holes abound.
- Pineland hosts the archeological site of the Calusa Indians, who occupied Pine Island for over 1,500 years, and the adjoining Randell Research Center. For boaters, there is a large marina offering fishing charters and an island taxi service to the outer islands of Useppa and Cayo Costa.
- Bokeelia has an abundance of tropical fruit and palm tree farms, an 18-hole golf course, fishing piers, a full-service marina, waterfront lodging, restaurants, and art gallery.
- Matlacha has evolved from a small fishing village to a center for local artists and writers, featuring an colorful mix of art galleries, boutiques and eateries. Still honoring its fishing heritage, it also hosts several fresh seafood shops and the "most fished off of" bridge in Florida!

Pine Island's easy, affordable access to Boca Grande Pass and the Gulf of Mexico offers some of the best fishing and boating in the world. Tarpon, snook, redfish, grouper, snapper, sea trout and many other varieties help make this a fisherman's paradise.

The waters of Matlacha Pass and the Pine Island Sound Aquatic Preserve are also delightful to explore by kayak or canoe.
